No, przeczytałem książkę dwie godziny does not work, because the aspect is perfective, and the direct object is książkę , not dwie godziny . You would not even use przez here. You would say Przeczytałem książkę w dwie godziny.
The only reason przesiedziałem w bibliotece dwie godziny works is that in this case, dwie godziny is in fact the direct object of przesiedziałem.

You can say “przeczytałem książkę w dwie godziny” - which means “I read a book in two hours”. If you skip “prze” and say “Czytałem książkę przez dwie godziny” it means “ I had been reading a book for two hours” sometime in the past.
