1. Refrain from any sexual activity (including masturbation) for at least 2 days and no more than 10 days. Longer or shorter periods of abstinence may result in a lower sperm count or decreased sperm motility.
2. A private room is available for semen collection within close proximity of our laboratory and is recommended for collection.
3. The semen specimen should be obtained by masturbation. Alternate methods may be discussed but are not recommended. Do not use any lubricant, including saliva, when collecting semen. Do not collect the specimen in a condom as these contain spermicidal agents, which will alter the results of the analysis. If a condom must be used, the laboratory will provide a special semen collection condom.
4. The specimen should be collected in a container provided by the ART lab. Be sure hands and penis are cleaned prior to collection. Avoid touching the inside of the cup. If any semen is spilled, DO NOT attempt to transfer it to the cup. Inform the lab personnel about the spill.
5. If the specimen was obtained outside of the collection room, bring the specimen to the laboratory within one hour after ejaculation. Do not expose the specimen to extremes of temperature. Place specimen container upright in a plastic bag, with the lid securely tightened, and keep specimen close to body temperature by transporting close to the body. The specimen should not be placed in a purse, pocket, or briefcase. Sperm do not have a long life outside of the body and at different temperatures. Delays in delivering semen and exposure to various temperatures will results in lower overall motile sperm count and poor semen cryopreservation.
6. Semen older than 1 hour will not be accepted for cryopreservation .
7. Please label the specimen collection container with the self-adhesive label printed with your name and registration number.
a. container is cracked, broken or leaking b. container not approved by the ART lab c. container is hot or cold to the touch d. specimen collected by use of condom not approved by the ART lab e. specimen brought in by unscheduled walk-in appointments.
9. One business day following the collection we will contact you about the quality and quantity of sperm in the test vial. Based on this information, we will make recommendations about the need for additional collections. We will also provide your primary oncologist with our recommendations, so medical treatment schedules can be made.

Semen collection refers to the process of obtaining semen from human males or other animals with the use of various methods, for the purposes of artificial insemination , or medical study (usually in fertility clinics ). Semen can be collected via masturbation [1] (e. g., from stallions [2] and canids [3] ), prostate massage , artificial vagina , penile vibratory stimulation (vibroejaculation) and electroejaculation . [4] Semen can be collected from endangered species for cryopreservation of genetic resources . [5]

Methods of semen collection from humans include:

The best specimen is produced when a short period of 3–5 days of abstinence is observed. More prolonged period does not yield better results. [8]

For semen collection from stallions, the most common method used is an artificial vagina; after collecting semen, it is tested, diluted, then stored according to the intended use. Semen can be either liquid or frozen. There are many kinds of preservatives used in dilution with semen - most contain energy and antibiotics for liquid, and protection from freezing for frozen semen. Many studies are ongoing to improve preservation, increase liquid semen storage time, and reduce or remove bacteria from semen. [12] [13] [14]

In order to collect semen from a male dog, an artificial vagina is prepared, [15] [16] which is a conical thin latex sleeve ending in a sterile collection tube. The inside of the latex sleeve is lightly lubricated. [17] The male is allowed to sniff a female in estrus. Experienced studs cooperate readily in the process. New studs often require encouragement in the form of manual stimulation, [18] [19] also known as "manual ejaculation". [20] [21] Generally the male will mount the female [ failed verification ] , and the collector quickly directs the dog's penis into the latex sleeve. The male ejaculates and the semen is collected in the tube. The semen is then drawn up into a long thin pipette. [22] [20] Prior to ejaculation, the penis is massaged inside its sheath. It is then extruded from its sheath, and the collector massages the dog's erect penis near the base of the bulbus glandis using the thumb and index finger. The dog begins pelvic thrusting movements at the onset of ejaculation. [23]

Semen can be collected from wolves via manual stimulation [24] or electroejaculation. [25] [26] [27]

It’s not uncommon to feel a bit uncomfortable about collecting a semen sample for sperm analysis. Many men describe the experience as awkward and even a little embarrassing. The conditions can be less than ideal: you’re performing “on demand,” most likely in an anonymous, sterile environment, with the clinic staff “knowing” that you’re doing something that’s normally private, and you’re handing it over once you’re done. Just keep in mind that it’s science. And it’s something that the nurses and staff at your clinic see multiple times a day, 365 days a year.
Masturbation is, most likely, the way you’ll collect your semen specimen. If you have severe male infertility resulting in few or no sperm, you may require a surgical procedure such as microsurgical epididymal sperm aspiration (MESA) or testicular sperm aspiration (TESA). Most doctors recommend you ejaculate directly into a provided sample cup and not use a condom. If you must use a condom, your lab will provide a special semen collection condom (that does not contain spermicide).
There are a few rules for collecting your semen sample:
If you’ve collected your sample outside of your clinic, you need to get it to the laboratory within one hour after ejaculation. Sperm does not have a long life outside of the body or in environments with fluctuating temperatures. Delays in delivering semen and exposure to various temperatures will result in lower overall motile sperm count and poor semen cryopreservation.
Your semen sample should be kept as close to body temperature as possible. The sperm motility value will be inaccurately low if the semen sample gets too cold or if it gets too hot.
Keep your specimen container upright in a plastic bag, with the lid securely tightened. The specimen should not be placed in any purse, pocket or briefcase.
Some clinics will require that you produce the sperm sample on site to make sure that you are able to produce a sample for a fertility cycle and to get the best possible sample.
Most likely you will be in a room that is meant for sample collection at your clinic. Here’s a head’s up: Most of these rooms (sometimes, it’s the clinic bathroom) are pretty, well . . . uninspiring. Just know this in advance. And you may want to bring your own porn, your smartphone, or a Victoria’s Secret catalog. By many accounts, the selection is often outdated.
