These alterations in lipid structure could be responsible for the inhibition of cholesterol biosynthesis in cells and apoptosis triggered by CBD treatment.

We observed that CBD rapidly and dose-dependently stimulated the death of apoptosis within SK-N-BE(2) cells, with about 50% of cells dying after 24 hours in the presence of 40 mM CBD (Fig. 5A). In addition, cotreatment of SK-N-BE(2) cells with CBD and the cholesterol biosynthesis inhibitor the drug atorvastatin, reduced apoptosis around twofold (Fig. 5B).

These results suggest that CBD hinders the homeostasis of cholesterol particularly in cells that are prone to mitochondrial dysfunction. This is also true for EHFD patients. This is in line with the findings of phosphoproteomics, which demonstrate that CBD treatment increases the AMP/ATP ratio and activates the AMPK kinase. Furthermore, Seahorse extracellular flux assays confirmed that CBD decreases the oxygen consumption rate in basal cells, consistent with the AMPK activation.
