ISRIB

Greeting, neuronerds! We are a group of chemists-pharmacologists, working on testing ISRIB on human. We have optimised synthesis nethods to get very high quality product. Also, we analyze every portion of newly synthsised isrib with a standart chemical analyzes like Nuclear Magnetic Resonance (H1, C13), Chromatography, Infrared spectroscopy and Mass spectroscopy. What is ISRIB?

ISRIB – is an inhibitor of low ISR (integrated stress response). The integrated stress response (ISR) protects cells from a variety of harmful stressors by temporarily halting protein synthesis. Every single cell is a very complex machine. For most of its functions, it synthesizes a wide variety of proteins. Stopping protein synthesis means stopping the functioning of the cell. In this case, the cell remains in limbo, and the functions performed by it are significantly reduced. Protein synthesis is a very hard and complex procces, so it can be regulated by different internal and external factors. One key in the protein synthesis is eukaryoitic initiation factor 2 (eIF2), wich can be phosphorylated by different stressors. If eIF2 is phosphorylated – protein synthesis stops. ISR mediate this posphorylation of eIF2. However, ISR can be activated by a huge variety of stressors: amino acid deprevation, endoplasmic reticulum stress, heme deprevation, prescence of heavy metals, viral infection, hypoxia and etc. After prolonged ISR, there can be triggered apoptosis (programmed cell death) in the cell. On the other hand, dephosphorylation of eIF-2 renews protein synthesis in the cell. ISR phosphorylation and dephosphorylation in the cell regulate normal cell life. 

There are a lot of problems in recovering brain after traumatic brain injury. The problem is that during a stroke, cells are put under stress. They are in limbo and due to the long-term functioning of the ISR, cells cannot return to normal functioning and this is the biggest problem during brain recovery after a stroke. ISRIB inhibits ISR and restores normal functioning of cells. eIF-2 plays a special role in the formation and consolidation of memory in the hippocampus. For normal memory formation, cells need good proteostasis. Inhibition of eIF-2 through mutations increased cognition in mice. Moreover, Isrib restores both short-term and long-term memory. ISRIB restores LTP (long term potentiation), increases cognitive abilities in both sick and healthy. 

So now we know what does mean ISR and we will return to ISRIB. Again, ISRIB inhibits low stress response, that is present in our everyday life. Interestingly, low activation of ISR is very toxic in lond term throughout the life. The integrated response to low stress is essentially the limiter of our energy potential. 1000 years ago, for example, when there was a lack of food, human was turned on to save energy. Now, in the conditions of the modern world, such function is not needed. ISRIB allows you to consume more food and this, for a minute, is an evolutionary breakthrough. All evolutionary breakouts have been associated with the ability to use more energy. This drug has very big therapeutic potential, therefore further research on humans is required.

Now I will try to tell you about the effects that were observed in our testers.

In about 15 minutes after intake (internasal) you start to feel some kind of ligntness in your head and body. Your thinking becomes faster and faster in geometrical progression, (visual) shapes of things become more recognizable, cornered. Tastes and smells start to be much more strong. 

In an hour the effects can bo compared to stims but the main difference is that this thing doesnt deal in any way with dopamine. You become some kind of u-human. Speaking about sleeping, 4-6 hrs/day is enough, you start to eat so much that your fridge will be shocked. Your cognitive and physical stats rise like never.
