Say there is a build with 50% penetration, and another with ~80% penetration, how does that translate to damage?
Would having over 100% penetration mean that monsters might as well have no elemental resistances,? (much like the inquisitor ascendancy?)
Penetration just lowers mobs' final resistance by the corresponding percentage. 100% doesn't mean resists are ignored, it means the mob's effective resistance is lowered by 100 (and into negative values). So the effect of penetration really depends on the mob's starting resists.
For example, if a mob has 75% resistance, then with 0 penetration you're doing 25% damage, with 50 penetration you're doing 75% damage (triple the damage of having no pen!), and with 80 pen you're dealing 105% damage (~4x the damage of no pen).
However, if a mob starts with 0 res, then 50% pen is just 50% more damage and 80% pen is 80% more.
[Your Damage] * [Monster Resistance - Your Penetration] = Damage Dealt.
It's a straight multiplier to damage. It can reduce resistances to any amount of negatives so having more is always good.

Think of your own resistances. When you have 50% resistance to an element, you take 50% less damage from that element. Resistances are very powerful, and therefore removing them is equally powerful. It essentially acts as a "more" multiplier to your damage. Also note that you can reduce enemy resistances by more than 100% and drop them into negative resists, at which point your damage is actually amplified against them.
Some things to consider when working with curses/resistance drops:
There are some mobs that can have over-capped resists, so if you reduce them by some amount, their "wasted" overcapped resist value will now come into play and cancel out some of your reductions.
Bosses have some level of reduced curse effect on them, making your elemental penetration curses less effective.
Just to add to the point about overcapped; if they have 200% fire resist, they're capped at 75%. If you penetrate 25%, this means you're doing 50% of base damage still - the amount they have over the cap doesn't matter.
This is what makes "penetrating resistance" better than "reducing resistance".
Curses generally reduce resistance, which makes the amount over cap matter - that is why penetration is such a strong star comparatively.
