BAT3
HLA-B associated transcript 3 (BAT3) is a gene that encodes a protein involved in protein degradation and apoptosis. It is also known as BAG6, BAG-6, or HLA-B associated transcript 3.
Bat3-deficient T cells display elevated expression of exhaustion markers, and knocking down Bat3 in myelin antigen-specific CD4 + T cells dramatically inhibits the development of experimental autoimmune encephalomyelitis while promoting the expansion of a dysfunctional Tim-3 hi IFN γlo CD4 + cell population.
Bat3-deficient T cells have elevated expression of exhaustion-associated molecules such as Tim-3, Lag3, Prdm1 and Pbx3, and Bat3 knockdown in myelin-antigen-specific CD4+ T cells markedly ...
DCs lacking BAT3, an adaptor protein that binds to TIM-3 on T cells, exhibited UPR overactivation, resulting in a tolerogenic phenotype that impaired antitumor T cell responses [14].
Here we show that human leukocyte antigen B (HLA-B)-associated transcript 3 (Bat3) binds to, and represses the function of, Tim-3. Bat3 protects T helper type 1 (TH1) cells from galectin-9-mediated cell death and promotes both proliferation and proinflammatory cytokine production.
Tim-3 adaptor protein Bat3 regulates T cell terminal differentiation and exhaustion in an mTORC2-dependent manner.
Because Bat3 is an adapter protein that binds to the cytoplasmic tail of Tim-3 and restrains its inhibitory function (12), therefore, it was tempting to hypothesize that Bat3 may function as an endogenous regulator of DCs function.
HLA-B-associated transcript 3 (Bat3) is identified as a binding partner to the Tim-3 intracellular tail that protects Th1 cells from galectin-9 mediated cell death, and promotes both proliferation and pro-inflammatory cytokine production. Interferon (IFN)γ-secreting Th1 CD4+ cells and Tc1 CD8+ cells play an essential role in protection against intracellular pathogens and viruses, and IFNγ ...
Here, we demonstrate that the adaptor molecule Bat3 acts as a molecular checkpoint of T cell exhaustion, with deficiency of Bat3 promoting a profound exhaustion phenotype, suppressing autoreactive T cell-mediated neuroinflammation.
Here, we demonstrate that the adaptor molecule Bat3 acts as a molecular checkpoint of T cell exhaustion, with deficiency of Bat3 promoting a profound exhaustion phenotype, suppressing autoreactive T cell-mediated neuroinflammation. Mechanistically, Bat3 acts as a critical mTORC2 inhibitor to suppress Akt function.

T cell exhaustion has been associated with poor prognosis in persistent viral infection and cancer. Conversely, in the context of autoimmunity, T cell exhaustion has been favorably correlated with long-term clinical outcome. Understanding the development of exhaustion in autoimmune settings may prov …

The methyltransferase DOT1L methylates histone H3 at K79 to facilitate specific biological events. H3K79 dimethylation (H3K79-2Me) by DOT1L influences the DNA damage response by promoting 53BP1 recruitment to DNA damage sites; however, it is unclear ...
Bat3 (HLA-B-associated transcript 3) was originally identified as a gene within the class III region of the human major histocompatibility complex on chromosome 6 (Spies et al. 1989; Banerji et al. 1990). The biological function of Bat3 is currently unknown. The Xenopus homolog of Bat3, Scythe, was recently shown to interact with Reaper (Rpr), a crucial regulator of apoptosis in Drosophila ...
Scythe/BAT3 is a member of the BAG protein family whose role in apoptosis has been extensively studied. However, since the developmental defects observed in Bat3-null mouse embryos cannot be explained solely by defects in apoptosis, we investigated whether ...
In response to DNA damage, p53 undergoes post-translational modifications (including acetylation) that are critical for its transcriptional activity. However, the mechanism by which p53 acetylation is regulated is still unclear. Here, we describe an essential role for HLA-B-associated transcript 3 ( …
The methyltransferase DOT1L methylates histone H3 at K79 to facilitate specific biological events. H3K79 dimethylation (H3K79‐2Me) by DOT1L influences the DNA damage response by promoting 53BP1 recruitment to DNA damage sites; however, it is unclear if this methylation is required as 53BP1 interacts with dimethylated H4 (H4K20‐2Me) with a much higher affinity. We demonstrate that H3K79 ...

The p53 pathway is a highly complex signaling network including several key regulators. HAUSP is a critical component of the p53 pathway acting as a deubiquitinase for both p53 and its key repressor Mdm2. Here, we identified a novel HAUSP-interacting protein, HLA-B-associated transcript 3 (Bat3) and found it to be capable of inducing p53 stabilization and activation via a HAUSP-dependent ...

HLA-B-associated transcript 3 (BAT3), also known as Scythe or BAG6, is a nuclear protein implicated in the control of apoptosis and natural killer (NK) cell-dendritic cell (DC) interaction. We demonstrate that BAT3 modulates the immune response by regulating the function of macrophages. BAT3 is released by macrophages in vitro and it down-regulates nitric oxide and proinflammatory cytokines ...

TNF receptor-associated factor 6 (TRAF6) plays a critical role in NF-κB and mitogen-activated protein kinase (MAPK) signaling pathways, both of which mediate macrophage activation in response to pathogen-associated molecular patterns such as bacterial endotoxin, lipopolysaccharides (LPS). In this study, we investigated whether HLA-B associated transcript-3 (BAT3) regulates LPS-induced ...
Background The human lymphocyte antigen (HLA) encoded BAT3/BAG6 recently attracted interest as a regulator of protein targeting and degradation, a function that could be exerted in the cytosol and in the nucleus. The BAT3 gene was described to consist of 25 exons. Diversity of transcripts can be generated by alternative RNA splicing, which may control subcellular distribution of BAT3 ...
