Everyone who has seen The Room remembers the bizarre moment when Johnny (Tommy Wiseau), seemingly out of nowhere, asks his best friend Mark "How's your sex life?"
The conversation begins with Johnny, who works at a bank, telling Mark that the bank won a big client. Mark asks who the client is. Johnny says he can't reveal the client's name, because it's confidential.
Mark presses him further for the name. Johnny, now quite annoyed, tells Mark "Anyway, how's your sex life?"
It's meant to be a sarcastic quip. It's like Johnny said "I can't give out that private information. Do you see me asking about your sex life? C'mon, man!"
The problem is that Tommy Wiseau is so terrible at delivering his lines that it sounds like he is seriously changing the subject from the bank to Mark's sex life. He reads the lines so monotone that you can't tell it's sarcasm.
That's the best explanation I can give. It makes even more sense if you subscribe to the fan theory (supported by Greg Sestero himself), that Mark is an undercover drug agent. Maybe that's why he's so interested in the new client.
If this were the only baffling thing about the movie I'd say you were onto something.
The only explanation you need is "Tommy Wiseau".
Explaining a movie like The Room... well I suppose it's good to have hobbies, even if they're a bit odd.

You know what's funny, I don't think the clip is on Youtube anymore, but there's something similar to what you're conveying in the "I did not hit her, I did NAHHHT!---oh, hi Mark!" scene.
I saw a clip where they were rehearsing, and what it's supposed to be is that Tommy is emotional and saying out loud "I did not hit her! It's not true! I did not hit her!" and crying, but then he see's Mark, and says "oh, hi Mark..." while scrambling to wipe away tears and not let on how upset he is.
As you can see...didn't exactly work out that way.
